What does glycine actually do for sleep?
It lowers core body temperature
Your core temperature naturally drops about 1 degree Celsius as you fall asleep, and a faster drop predicts faster sleep onset. Orally ingested glycine amplifies this signal: it crosses into circulation, raises glycine levels in cerebrospinal fluid, and increases cutaneous blood flow (peripheral vasodilation), dumping heat through the skin. In a 2015 Neuropsychopharmacology study in rats, this effect was traced to NMDA receptors in the suprachiasmatic nucleus (SCN) - the brain's master clock. Blocking NMDA receptors, or removing the SCN, abolished both the temperature drop and the sleep-promoting effect. So glycine is not a sedative; it nudges a circadian mechanism.
What the human trials found
In the Yamadera 2007 polysomnography study, 11 volunteers with chronically unsatisfactory sleep took 3 g of glycine or placebo before bed. Glycine improved subjective sleep quality and sleep efficiency, and shortened both the objective time to fall asleep and the time to reach slow-wave sleep - without distorting overall sleep architecture. The honest caveats
These are consistent results, but the entire human evidence base is 3 trials totaling under 30 participants, with authors affiliated with or funded by Ajinomoto - a company that sells glycine. That does not make the data wrong; industry-run pilot studies are how much nutrition science starts. It does mean the effect size is uncertain and has never been confirmed by a large independent replication. Set expectations at "modest and subtle," not "knocked out." The math: magnesium glycinate is not a glycine supplement
A common assumption is that magnesium glycinate covers both bases. Run the numbers: pure magnesium bisglycinate is roughly 14% magnesium and 86% glycine by weight. A serving providing 200 mg of elemental magnesium therefore carries only about 1.2 g of glycine - well under half the 3 g dose used in the sleep trials. To reach 3 g of glycine from magnesium bisglycinate alone you would need about 490 mg of elemental magnesium, far above the 350 mg/day tolerable upper limit for supplemental magnesium set by the NIH. Many "glycinate" products are also buffered blends cut with magnesium oxide, containing even less glycine. So the glycine in your magnesium glycinate is a sub-hypnotic dose - if you want the studied glycine effect, add plain glycine powder.
Where the magnesium evidence stands
Magnesium's sleep data is arguably weaker than glycine's, just larger. The 2021 systematic review and meta-analysis pooled trials in older adults and found sleep-onset latency about 17 minutes shorter versus placebo - but graded the evidence low to very low quality. The best single trial (Abbasi 2012, 46 elderly insomniacs, 500 mg for 8 weeks) improved insomnia severity, sleep time, and sleep efficiency - in a population likely to be magnesium-deficient. Is glycine safe? What are the side effects?
